目的 :研究亚甲蓝 /光化学灭活病毒技术对人血浆中抗体活性的影响。方法 :将不同浓度的亚甲蓝分别加入含有特种抗体的血浆中 ,经JY I型血浆病毒灭活仪处理后取样 ,用ELISA等方法检测血浆中的抗体活性。结果 :在光照度 4 0 0 0 0lx条件下 ,经 1μmol/L亚甲蓝作用 30min ,人血浆中的抗体活性未见明显改变 ,但随着亚甲蓝浓度的升高和光照时间的延长 ,血浆中的抗体活性略有下降。结论 :本研究的亚甲蓝 /光化学灭活病毒技术对血浆中的抗体活性影响较小 ,可用该技术对康复期的SARS患者血浆进行病毒灭活处理 ,保证该血浆临床应用的安全性。
Objective: To study the effect of methylene blue / photochemical inactivated virus on the activity of human plasma antibody. Methods: Different concentrations of methylene blue were added to the plasma containing special antibodies, respectively. After treatment with JY I plasma virus inactivator, the samples were taken and the plasma antibody activity was measured by ELISA. Results: Under the illumination of 40 000 lx, the activity of the antibody in human plasma did not change significantly after treated with 1 μmol / L methylene blue for 30 min. However, with the increase of methylene blue concentration and illumination time, plasma The antibody activity in the slightly decreased. Conclusion: The methylene blue / photochemical inactivated virus technology in this study has little effect on the antibody activity in plasma. This technique can be used to inactivate the plasma of convalescent SARS patients and ensure the safety of the plasma for clinical application.
