目的 观察保肝类中药复方防治CCl4 所致大鼠肝纤维化模型的疗效 ,并探讨其作用机制。 方法 整体实验 ,采用常规切片、HE、Mallory染色和免疫组织化学ABC法检测Ⅲ型胶原和层黏连蛋白 ;离体实验 ,采用肝星形细胞分离培养 ,检测Ⅲ型胶原和层黏连蛋白 ,并经图像分析系统作定量分析。 结果 保肝类中药复方能减少肝血窦及纤维隔内肌样成纤维细胞的数量 ,抑制纤维隔的纤维组织增生 ,并抑制活化的肝星形细胞分泌Ⅲ型胶原和层黏连蛋白。 结论 保肝类中药复方防治肝纤维化实验疗效明显 ,其抗肝纤维化作用机理与抑制细胞外基质的合成和促进其降解有关。
Objective To observe the curative effect of liver-protective Chinese herbal compound on prevention and treatment of liver fibrosis induced by CCl4 in rats, and to explore its mechanism of action. Methods The whole experiment was performed using conventional sectioning, HE, Mallory staining and immunohistochemical ABC methods to detect type III collagen and laminin; in vitro experiments, hepatic stellate cells were isolated and cultured to detect type III collagen and laminin. And quantitative analysis by the image analysis system. Results Hepatoprotective Chinese herbal compound can reduce the number of myofibroblasts in hepatic sinusoids and fibrous septa, inhibit fibrous tissue proliferation in fibrous septa, and inhibit activated hepatic stellate cells from secreting type III collagen and laminin. Conclusion The protective effect of hepatoprotective Chinese herbal compound on hepatic fibrosis is obvious, and its anti-hepatic fibrosis mechanism is related to inhibiting the synthesis of extracellular matrix and promoting its degradation.
