为了探索蛹虫草液体发酵中适合虫草素积累的培养基和发酵培养条件,以半定量薄层色谱法为检测手段,以虫草素含量为指标,通过单因素试验及正交试验优选的方法,最终优选出的培养基配方为蛋白胨15 g.L-1、葡萄糖20 g.L-1、磷酸二氢钾1 g.L-1、七水合硫酸镁0.5 g.L-1、维生素B1 10 mg.L-1、萘乙酸2 mg.L-1。优选出的培养条件为发酵液pH7,培养温度25℃,光照时间12 h。
In order to explore the culture medium and fermentation conditions suitable for the accumulation of cordycepin in the liquid fermentation of Cordyceps militaris, semi-quantitative TLC was used as the detection method, and the content of cordycepin was taken as the index. Through the single factor experiment and the orthogonal experiment, The optimal medium was peptone 15 gL-1, glucose 20 gL-1, potassium dihydrogen phosphate 1 gL-1, magnesium sulfate heptahydrate 0.5 gL-1, vitamin B1 10 mg.L-1, naphthalene acetic acid 2 mg .L-1. The optimal culture conditions were fermentation broth pH7, culture temperature 25 ℃, illumination time 12 h.
