目的探讨Raptor调节前列腺癌上皮-间质转化(EMT)作用及机制研究。方法构建Raptor-shRNA慢病毒载体并筛选具有稳定转染Raptor-shRNA DU145细胞株,应用transwell观察沉默Raptor后DU145细胞株侵袭力的变化,Western Blot观察沉默Raptor后DU145细胞E-cadherin,β-catenin,N-cadherin和vimentin表达,以及RhoA活性表达。结果成功构建Raptor-shRNA DU145细胞株;transwell检测稳定转染Raptor-shRNA后DU145细胞侵袭力下降(P<0.01);Western Blot检测显示该细胞株E-cadherin和β-catenin表达下降,N-cadherin和vimentin表达升高,RhoA活性表达下降。结论 Raptor与前列腺癌细胞EMT的发生有关,其作用可能是通过RhoA信号途径。
Objective To investigate the role and mechanism of Raptor in the regulation of epithelial-mesenchymal transition (EMT) in prostate cancer. Methods The Raptor-shRNA lentiviral vector was constructed and transfected into Raptor-shRNA DU145 cells. The transwell was used to observe the invasiveness of DU145 cells after silencing Raptor. Western Blot was used to observe the expression of E-cadherin and β-catenin , N-cadherin and vimentin expression, and RhoA activity. Results The Raptor-shRNA DU145 cell line was constructed successfully. The invasiveness of DU145 cells was down-regulated by transwell assay (P <0.01), and the expression of E-cadherin and β-catenin was down-regulated by Western Blot. And vimentin expression increased, RhoA activity decreased. Conclusion Raptor is associated with the occurrence of EMT in prostate cancer cells, and its role may be through the RhoA signaling pathway.
