目的:探讨siRNA下调三重基序(tripartite motif,TRIM)46蛋白表达对结直肠癌HCT116细胞增殖和侵袭功能的影响及可能的作用机制。方法:将TRIM46 siRNA转染至人结直肠癌HCT116细胞后,应用实时荧光定量PCR和蛋白质印迹法检测细胞中TRIM46 mRNA及蛋白的表达水平,CCK-8法和Transwell法分别检测下调TRIM46表达对HCT116细胞增殖和侵袭能力的影响,蛋白质印迹法检测细胞中SH2结构域酪氨酸磷酸酶1[Src homology 2(SH2)-containing tyrosine phosphatase 1,SHP1)]蛋白的表达。结果:TRIM46 siRNA转染后,HCT116细胞中TRIM46 mRNA及蛋白表达水平被显著抑制(P值均<0.05)。下调TRIM46表达可显著抑制结直肠癌HCT116细胞的增殖和侵袭(P值均<0.05),而SHP1蛋白表达水平明显升高(P<0.05)。结论:泛素连接酶TRIM46可能通过抑制SHP1表达,促进结直肠癌细胞的增殖和侵袭。
Objective: To investigate the effect of down-regulation of TRIM 46 protein on the proliferation and invasion of human colorectal cancer HCT116 cells and its possible mechanism. Methods: TRIM46 siRNA was transfected into human colorectal cancer cell line HCT116. The expression of TRIM46 mRNA and protein was detected by real-time fluorescence quantitative PCR and Western blotting. The expression of TRIM46 mRNA and protein was detected by Transfection assay and CCK-8 respectively. (SH2) -containing tyrosine phosphatase 1 (SHP1)] protein in SH2 cells was detected by Western blotting. Results: After TRIM46 siRNA transfection, the expression of TRIM46 mRNA and protein in HCT116 cells was significantly inhibited (all P <0.05). Down-regulation TRIM46 expression significantly inhibited the proliferation and invasion of colorectal cancer HCT116 cells (all P <0.05), while the SHP1 protein expression was significantly increased (P <0.05). Conclusion: Ubiquitin ligase TRIM46 may promote the proliferation and invasion of colorectal cancer cells by inhibiting SHP1 expression.
