目的:研究艳山姜挥发油对氧化低密度脂蛋白(ox-LDL)诱导人脐静脉内皮细胞损伤的保护作用及一氧化氮合酶-一氧化氮(NOS-NO)的影响。方法:体外传代培养人脐静脉内皮细胞,给予0.1μg·mL-1和0.01μg·mL-1艳山姜挥发油孵育24h后,加入100μg·mL-1 ox-LDL作用24h诱导人脐静脉内皮细胞损伤模型,倒置相差显微镜及吉姆萨染色观察细胞形态学变化,比色法分析细胞培养液NO的含量与细胞裂解液NOS分型活力。结果:Ox-LDL与人脐静脉内皮细胞作用24h后,细胞胞膜皱缩,细胞数目减少,tNOS与eNOS活力显著降低,培养液NO含量降低。艳山姜挥发油显著改善细胞形态学变化,提高细胞tNOS与eNOS活力及培养液中NO含量。模型组与艳山姜挥发油组均对iNOS未见显著影响。结论:艳山姜挥发油对ox-LDL诱导人脐静脉内皮细胞损伤具有保护作用,其保护作用与改善eNOS活力,提高NO水平相关。
AIM: To investigate the protective effect of volatile oil from Yammycinmus pilosula on oxidative low density lipoprotein (ox-LDL) -induced injury of human umbilical vein endothelial cells and the effect of nitric oxide synthase (NO) on it. Methods: Human umbilical vein endothelial cells (ECs) were subcultured in vitro and incubated with 100 μg · mL-1 ox-LDL for 24 h after being treated with 0.1 μg · mL-1 and 0.01 μg · mL-1 YPC respectively. The morphological changes of the cells were observed by inverted phase contrast microscope and Giemsa staining. The content of NO in cell culture medium and the activity of NOS typing in cell lysate were analyzed by colorimetric assay. Results: Ox-LDL and human umbilical vein endothelial cells for 24 h, cell membrane shrinkage, the number of cells decreased, tNOS and eNOS activity was significantly reduced, NO content in the culture fluid decreased. Yam Ginger volatile oil significantly improved cell morphology changes, increase cell activity of tNOS and eNOS and NO in culture medium. The model group and the yam ginger volatile oil group had no significant effect on iNOS. CONCLUSION: Volatile oil from Yammycin Ginseng has a protective effect on ox-LDL-induced injury of human umbilical vein endothelial cells, and its protective effect is related to improving eNOS activity and increasing NO level.
