目的分析国内不同产地蒺藜果实之间以及果实与全草之间化学成分的差异。方法色谱分离采用Acquity HSS T3C18色谱柱(Waters,100 mm×2.1 mm,1.8μm),0.1%甲酸水溶液-乙腈为流动相进行梯度洗脱,质谱分析利用四级杆串联飞行时间质谱(Q-TOF MSE);所得数据采用主成分分析法(PCA)及正交偏最小二乘判别分析(OPLS-DA)进行处理,差异标志物通过精确相对分子质量和碎片离子鉴定结构。结果 PCA结果显示本实验所研究的各产地蒺藜果实之间无显著差异,而蒺藜果实与全草之间存在一定差异性,并利用液质联用技术鉴定了两者之间差异性较大的12个化学成分,其中9个主要存在于果实中,另外3个主要存在于全草中。结论通过研究国内几个重要产区蒺藜果实之间及果实与全草之间化学成分的差异,为蒺藜资源的合理利用以及临床用药提供理论依据。
Objective To analyze the differences of the chemical composition between the fruits of Tribulus terrestris and the whole plant in China. Methods The chromatographic separation was performed on a Waters Acquity HSS T3C18 column (Waters, 100 mm × 2.1 mm, 1.8 μm) with gradient elution with 0.1% formic acid in acetonitrile as the mobile phase. The mass spectra were analyzed by quadrupole tandem time of flight mass spectrometry (Q-TOF MSE). The data were processed by Principal Component Analysis (PCA) and Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA). Differential markers were identified by accurate relative molecular mass and fragment ions. Results PCA results showed that there was no significant difference between the fruits of Tribulus terrestris studied in this study, while there was some difference between Tribulus terrestris fruits and whole plant, and the difference between the two methods was identified by LC-MS Twelve chemical constituents, of which nine are found mainly in the fruit and the other three mainly in the whole plant. Conclusion By studying the differences in the chemical composition between the fruits of Tribulus terrestris and the whole plant in several important producing areas in China, this study provides a theoretical basis for the rational utilization of Tribulus terrestris resources and clinical drug use.
