目的建立高同型半胱氨酸血症大鼠模型,观察高同型半胱氨酸血症对血脑屏障的影响。方法36只SD大鼠分为对照和模型(皮下注射蛋氨酸)两组。采用高压液相色谱法检测同型半胱氨酸(homocysteine,Hcy)浓度;比色法检测大鼠大脑伊文思蓝含量观测血脑屏障的通透性改变;透射电镜观察大脑皮质毛细血管形态改变。结果对照组大鼠在不同时间点血浆Hcy水平和脑组织伊文思蓝含量均无明显变化,血脑屏障结构亦正常。模型组大鼠血浆Hcy水平和脑组织伊文思蓝含量在第4w和第8w均显著增高,同时血脑屏障也呈现不同程度的破坏。结论高同型半胱氨酸血症可损伤大鼠血脑屏障的结构和功能。
Objective To establish a rat model of hyperhomocysteinemia and observe the effect of hyperhomocysteinemia on the blood-brain barrier. Methods Thirty-six SD rats were divided into control group and model group (subcutaneous methionine injection). The concentration of homocysteine (Hcy) was detected by high pressure liquid chromatography. The permeability of blood brain barrier was observed by using the Evans blue content of brain in rats by colorimetry. The morphological changes of capillaries were observed by transmission electron microscope. Results There was no significant change in plasma Hcy levels and brain Evans blue content in control rats at different time points, and the structure of blood-brain barrier was also normal. The level of Hcy in plasma and the content of Evans blue in brain tissue in model group were significantly increased at 4w and 8w, meanwhile, the blood-brain barrier also showed different degrees of damage. Conclusion Hyperhomocysteinemia can damage the structure and function of BBB in rats.
