目的研究盐敏感性高血压大鼠肾损害中Th17细胞的表达以及吗替麦考酚酯(MMF)对高血压肾损害的作用。方法 24只健康雄性Wistar大鼠用BP-6A无创血压仪每周监测大鼠血压1次,8周后处死大鼠,比色法检测大鼠24h尿蛋白排泄水平的变化,流式细胞仪检测外周血Th17细胞占CD4+T细胞的比例;酶联免疫吸附法检测血清中白细胞介素-17(interleukin-17,IL-17)水平。结果模型组血压、尿蛋白排泄水平、Th17细胞的比例、IL-17水平与对照组相比均升高(P<0.01)。与模型组比较,MMF组血压、尿蛋白排泄水平、Th17细胞的比例、IL-17水平均降低(P<0.01)。结论 Th17细胞表达上调可能参与了盐敏感性高血压肾损害的过程,MMF可以减轻Th17细胞在肾脏的浸润。
Objective To investigate the expression of Th17 cells and the effect of mycophenolate mofetil (MMF) on renal damage in hypertensive rats with salt-sensitive hypertension. Methods Twenty-four healthy male Wistar rats were anesthetized once a week with BP-6A non-invasive blood pressure monitor. Rats were sacrificed after 8 weeks. Urinary protein excretion levels were measured by colorimetric assay. Flow cytometry Th17 cells in peripheral blood accounted for the proportion of CD4 + T cells; serum interleukin-17 (IL-17) levels by enzyme-linked immunosorbent assay. Results The levels of blood pressure, urinary protein excretion, the ratio of Th17 cells and IL-17 in model group were higher than those in control group (P <0.01). Compared with model group, blood pressure, urinary protein excretion, Th17 cell ratio and IL-17 level in MMF group were decreased (P <0.01). Conclusion Upregulation of Th17 cells may be involved in the process of salt-induced hypertensive renal damage. MMF can reduce the infiltration of Th17 cells in the kidney.
