目的:研究何首乌的2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(TSG)对实验性高脂血症大鼠的调脂作用。方法:高脂血症大鼠分别给予TSG 90,180mg.kg-1,阿托伐他汀钙10mg.kg-1和生理盐水2mL灌胃,每日1次,连续给药1周,同时设正常对照组。检测给药前后血清总胆固醇(TC)、总甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)含量,并计算动脉粥样硬化指数;检测给药后各组低密度脂蛋白受体(LDLR)和羟甲戊二酰辅酶A受体mRNA的表达量。结果:TSG给药1周,能够有效控制高脂血症模型大鼠血清TC和LDL-C升高,可显著降低动脉粥样硬化指数,并且能够增加LDLR的表达。结论:TSG具有良好的调节血脂作用,是中药何首乌调节血脂作用的主要活性成分,具有显著降低血清TC,LDL-C和AI的作用。其中,TSG对于LDL-C和AI的调节作用,以及对基因表达的影响为首次报道。
Objective: To study the effect of 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside (TSG) from Polygonum multiflorum Thunb on lipid-regulating rats with experimental hyperlipidemia. METHODS: Rats with hyperlipidemia were given TSG 90, 180 mg.kg-1, atorvastatin calcium 10 mg.kg-1, and normal saline 2 mL, once a day, for 1 week, and normal control. group. Serum total cholesterol (TC), total triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high-density lipoprotein cholesterol (HDL-C) levels were measured before and after administration, and the atherosclerosis index was calculated. The expression levels of low-density lipoprotein receptor (LDLR) and hydroxyglutaryl-coenzyme A receptor mRNA were measured in each group after administration. RESULTS: TSG administration for 1 week could effectively control the serum levels of TC and LDL-C in hyperlipidemia model rats, significantly reduce the atherosclerosis index, and increase the expression of LDLR. Conclusion: TSG has a good role in regulating blood lipids, and it is the main active ingredient in the regulation of blood lipids by the Chinese medicine Polygonum multiflorum. It has a significant effect on reducing serum TC, LDL-C and AI. Among them, the regulatory effect of TSG on LDL-C and AI, as well as the effect on gene expression, was reported for the first time.
