目的:建立高效液相色谱法测定地胆草中4,5-二咖啡酰奎宁酸含量的方法。方法:色谱柱为Agilent SB C18柱(250mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸溶液(17∶83);流速为1.0 ml·min-1;检测波长为327 nm;柱温为30℃,进样量10μl。结果:4,5-二咖啡酰奎宁酸在0.023 5~2.352 0μg范围内线性关系良好(r=0.999 9),平均加样回收率为98.75%,RSD为1.24%(n=6)。结论:所用方法简便、快速、准确,可作为测定地胆草中4,5-二咖啡酰奎宁酸含量的方法。
Objective: To establish a HPLC method for the determination of 4,5-dicaffeoylquinic acid in Gentian. METHODS: The column was an Agilent SB C18 column (250 mm × 4.6 mm, 5 μm) with a mobile phase of acetonitrile-0.1% phosphoric acid solution (17:83), a flow rate of 1.0 ml · min -1 and a detection wavelength of 327 nm. 30 ℃, injection volume 10μl. Results: The linearity of 4,5-dicaffeoylquinic acid in the range of 0.023 5 ~ 2.352 0μg was good (r = 0.999 9). The average recovery was 98.75% and the RSD was 1.24% (n = 6). Conclusion: The method is simple, rapid and accurate and can be used as a method for the determination of 4,5-dicaffeoylquinic acid in Gentian.
