目的:研究植物激素对固体培养丹参毛状根生长和丹参酮ⅡA生物合成的影响。方法:组织培养法诱导毛状根、超声法提取丹参酮、高效液相色谱法测定丹参酮。结果:在1/2 MS培养基中添加一定浓度GA3与6-BA的组合激素对丹参毛状根生长有抑制作用,最高抑制作用达到100%(毛状根死亡),而添加1.0 mg/L KT与1.0mg/L IBA的组合激素则可以明显促进丹参毛状根的生长,生长量是对照的3.251倍;添加0.2 mg/L NAA和3.0mg/L 6-BA与3.0 mg/L NAA的组合激素时丹参毛状根体内丹参酮ⅡA的总含量最高;添加2.0 mg/L 6-BA则对丹参酮ⅡA的生物合成有显著促进作用,丹参酮ⅡA浓度是对照的3.012倍。结论:在丹参毛状根培养基中加入不同的植物激素及其组合激素对毛状根的生长和丹参酮ⅡA的生物合成有显著的影响。
Objective: To study the effects of plant hormones on the growth of hairy root and the biosynthesis of tanshinone ⅡA in solid cultured Salvia miltiorrhiza. Methods: Tissue culture was used to induce the hairy roots. Tanshinone was extracted by ultrasonic method and tanshinone was determined by high performance liquid chromatography. Results: The combination of GA3 and 6-BA in 1/2 MS medium inhibited the growth of hairy root of Salvia miltiorrhiza Bunge. The maximum inhibitory effect reached 100% (the hairy root died), while the addition of 1.0 mg / L The combination of KT and 1.0mg / L IBA could obviously promote hairy root growth of Salvia miltiorrhiza Bunge, which was 3.251 times of that of the control. Adding 0.2mg / L NAA and 3.0mg / L 6-BA with 3.0mg / L NAA The total content of tanshinoneⅡA in the hairy root of Salvia miltiorrhiza Bunge was the highest when combined with hormone. The addition of 2.0 mg / L 6-BA promoted the tanshinoneⅡA biosynthesis significantly, and the concentration of tanshinone ⅡA was 3.012 times of the control. CONCLUSION: The addition of different plant hormones and their combination hormones to the hairy root of Salvia miltiorrhiza hair-rooted medium has a significant effect on the growth of hairy roots and the biosynthesis of tanshinone IIA.
