目的:观察单抗PF18-3对小鼠胸腺树突状细胞系(MTSC4)增强胸腺细胞凋亡作用的影响并对PF18-3识别分子进行特征分析。方法:用DNA梯度断裂片段电泳检测PF18-3对凋亡过程的抑制作用;用膜蛋白斑点杂交和免疫共沉淀对其识别分子进行特征分析。结果:PF18-3抑制MTSC4对胸腺细胞凋亡的增强作用。其识别分子为分子量31000的膜蛋白,在不同胸腺基质细胞系分布不同,与其凋亡介导增强作用一致。结论:PF18-3识别分子参与凋亡介导增强过程,为非凋之介导分子CD95/配体(Fas/FasL)分子,值得进一步研究。
OBJECTIVE: To observe the effect of monoclonal antibody PF18-3 on thymocyte apoptosis in mouse thymic dendritic cell line (MTSC4) and to characterize PF18-3 recognition molecule. Methods: The inhibitory effect of PF18-3 on apoptosis was detected by DNA gradient fragment electrophoresis. The characteristics of the recognition molecules were analyzed by dot blot hybridization and co-immunoprecipitation. Results: PF18-3 inhibited the enhancement of MTSC4 on thymocyte apoptosis. Its molecular recognition molecular weight of 31000 membrane protein, in different thymic stromal cell line distribution, consistent with the role of apoptosis-mediated enhancement. Conclusion: The PF18-3 recognition molecule is involved in the process of apoptosis-mediated enhancement. It is a non-apoptotic CD95 / ligand (Fas / FasL) molecule that deserves further study.
