用三种抗原加工阻断剂预处理抗原提呈细胞(APC)1h后再用天花粉蛋白(Tk)脉冲处理,观察经由APC递呈的Tk对PMA和A23187诱发T细胞增殖的作用。结果表明,这些药物均能不同程度地阻断Tk的抑制效应,以氯喹和Leupeptin的作用更为明显。用胶体金标记Tk(Tk-G),电镜下观察Tk在T细胞和APC内的定位,发现Tk-G仅与APC发生细胞表面粘附,继而内化,先后出现在APC的内体和溶酶体中。上述实验提示,在本实验采用的剂量范围内,Tk可通过外源性抗原的加工途径,被APC处理后,调节T细胞的免疫应答。
The antigen-presenting cells (APCs) were pretreated with three antigen processing blockers for 1 h and pulsed with Trichosanthin (Tk) to observe the effect of Tk presented by APC on the proliferation of T cells induced by PMA and A23187. The results showed that these drugs can block the inhibitory effect of Tk to varying degrees, and the effect of chloroquine and Leupeptin is more obvious. Tk (Tk-G) was labeled with colloidal gold. The localization of Tk in T cells and APC was observed under electron microscope. It was found that Tk-G only adhered to the cell surface of APC, followed by internalization, and appeared in the endosomes of APC. Enzyme body. The above experiments suggest that within the dose range used in this experiment, Tk can be treated by APC through the processing route of exogenous antigen and regulate the immune response of T cells.
