目的:观察清热化瘀方联合缺氧预处理(HP)骨髓间充质干细胞(BMSCs)移植对脑缺血损伤大鼠血管新生标志物CD31表达变化及神经功能的影响。方法:将120只大鼠随机为6组:假手术组(SO组)、模型组(MCAO组)、BMSCs移植对照组(BMSCs组)、HP-BMSCs移植组(HP-BMSCs组)、BMSCs移植联合清热化瘀方组(中药对照组)以及HP-BMSCs移植联合清热化瘀方组(中药干预组),各组根据取材时间点又分为7天、14天两个亚组,每个亚组10只大鼠。线栓法建立大鼠大脑中动脉梗阻模型,根据分组情况给予清热化瘀颗粒灌胃及经尾静脉移植BMSCs或HP-BMSCs,参照m NSS评分法进行神经功能评分,采用免疫组化染色法检测血小板-内皮细胞黏附分子(CD31)及Brdu标记的BMSCs细胞。结果:与MCAO组比较,中药对照组、中药干预组、HP-BMSCs组、BMSCs组神经功能评分显著减少,差异均有统计学意义(P<0.05,P<0.01)。与SO组比较,其它各组CD31表达均显著上调,中药干预组上调的最为明显,差异均有统计学意义(P<0.01)。与HP-BMSCs组比较,BMSCs组神经功能评分增加,CD31表达下降,Brdu标记的阳性细胞数较低;中药干预组神经功能评分显著减少,CD31表达明显上调,Brdu标记的阳性细胞数显著增多,差异均有统计学意义(P<0.05,P<0.01)。与中药对照组比较,中药干预组神经功能评分明显减少,CD31表达量显著增加,Brdu标记的阳性细胞数显著增多,差异均有统计学意义(P<0.05,P<0.01)。结论:清热化瘀方联合HP-BMSCs移植可改善脑缺血损伤SD大鼠的神经功能,可能和其促进大鼠脑缺血区血管新生标志物CD31的表达相关。
Objective: To observe the effect of Qingre Huayu Recipe combined with hypoxic preconditioning (BMSCs) transplantation on the expression of angiogenesis marker CD31 and neurological function after cerebral ischemia in rats. Methods: 120 rats were randomly divided into 6 groups: sham operation group (SO group), model group (MCAO group), BMSCs transplantation control group (BMSCs group), HP-BMSCs transplantation group Combined with Qingre Huayu Recipe (traditional Chinese medicine control group) and HP-BMSCs transplantation combined with Qingre Huayu Recipe group (traditional Chinese medicine intervention group), each group was divided into 7 days and 14 days in two subgroups, each sub-group Group 10 rats. The model of middle cerebral artery occlusion in rats was established by thread occlusion. BMSCs or HP-BMSCs were transplanted via the Qingre Huayu granules and tail vein through grouping, and the neurological function score was evaluated by m NSS score method. Immunohistochemical staining Platelet - endothelial cell adhesion molecule (CD31) and Brdu - labeled BMSCs. Results: Compared with MCAO group, the scores of neurological function in TCM control group, traditional Chinese medicine intervention group, HP-BMSCs group and BMSCs group were significantly decreased, the difference was statistically significant (P <0.05, P <0.01). Compared with the SO group, the expression of CD31 in other groups were significantly increased, the most significant up-regulation in the intervention group of traditional Chinese medicine, the difference was statistically significant (P <0.01). Compared with the HP-BMSCs group, the neurological score increased, the expression of CD31 decreased and the number of Brdu-labeled positive cells decreased in the BMSCs group. The neurological scores, the expression of CD31 and the number of Brdu-labeled positive cells in the BMSCs group were significantly decreased The differences were statistically significant (P <0.05, P <0.01). Compared with the traditional Chinese medicine control group, the scores of neurological function decreased significantly, the expression of CD31 increased significantly, and the number of Brdu labeled positive cells increased significantly. The difference was statistically significant (P <0.05, P <0.01). Conclusion: Qingre Huayu Recipe combined with HP-BMSCs transplantation can improve the neurological function of SD rats with cerebral ischemic injury, which may be related to its role in promoting the expression of angiogenesis marker CD31 in cerebral ischemic area.
