目的探讨促红细胞生成素(EPO)对糖尿病大鼠心肌纤维化及心功能的影响。方法采用链脲佐菌素一次性腹腔注射成功建立糖尿病动物模型雄性SD大鼠20只,随机均分为模型对照(A)组和EPO处理(B)组;另取10只SD大鼠作为空白对照(C)组。12周后,称取心脏重量和大鼠体重,计算心体比;Western blot检测心肌组织转化生长因子β(TGF-β)表达;天狼猩红染色测心肌组织胶原含量;心脏超声检测心功能。结果与C组比较,A组心肌组织TGF-β蛋白表达和胶原含量明显增加(P<0.01),心体比、左室舒张末期内径(LVDd)和左室收缩末期内径(LVDs)明显增加(P<0.05),射血分数(EF)明显下降(P<0.01)。与A组比较,B组心肌组织TGF-β蛋白表达及胶原含量均明显减少(P<0.01),EF值增加(P<0.05),LVD d和LVDs明显减少(P<0.05)。结论EPO通过减少糖尿病大鼠TGF-β的表达来逆转心肌间质纤维化,改善心功能。
Objective To investigate the effect of erythropoietin (EPO) on myocardial fibrosis and cardiac function in diabetic rats. Methods A total of 20 male SD rats with diabetes mellitus were successfully established by intraperitoneal injection of streptozotocin (STZ). The rats were randomly divided into model group (A) and EPO group (B). Another 10 SD rats were used as blank Control (group C). Twelve weeks later, the heart weight and body weight were weighed and the ratio of heart to heart was calculated. The expression of TGF-β in myocardial tissue was measured by Western blot. The content of collagen in myocardium was detected by Sirius red staining. Cardiac function was detected by echocardiography. Results Compared with group C, the expression of TGF-βprotein and collagen in myocardium of group A were significantly increased (P <0.01), and the ratio of cardiocyte to heart, end-diastolic diameter (LVDd) and end-diastolic diameter P <0.05), ejection fraction (EF) decreased significantly (P <0.01). Compared with group A, the expression of TGF-β and the content of collagen in group B were significantly decreased (P <0.01), the value of EF was increased (P <0.05), and LVD d and LVDs were significantly decreased (P <0.05). Conclusion EPO can reverse myocardial interstitial fibrosis and improve cardiac function by reducing TGF-β expression in diabetic rats.
