目的:检测9-顺维甲酸(9-cis-RA)处理前后体外培养的人肺癌组织及其对照组织细胞周期蛋白因子CyclinD1、细胞周期蛋白依赖性激酶Cdk4、细胞周期蛋白酶抑制因子P16转录水平的变化。探讨其抑制肺癌组织细胞生长的作用机制。方法:采用半定量逆转录聚合酶链式反应(RT-PCR)分析CyclinD1、Cdk4、P16转录的变化。结果:9-cis-RA既可降低体外培养的人肺癌组织Cdk4的转录,又可升高其P16的转录。对肺癌组织和正常肺组织CyclinD1的转录的影响无统计学意义。对正常肺组织Cdk4、P16的转录的影响也无统计学意义。结论:9-cis-RA抑制肺癌组织细胞增殖的作用机理与其调节Cdk4、P16的转录有关。
OBJECTIVE: To detect the transcriptional levels of cyclinD1, cyclin-dependent kinase Cdk4, and cyclin-inducible factor P16 in human lung cancer tissues and their control tissues before and after 9-cis-RA treatment Variety. Methods: The transcription of CyclinD1, Cdk4 and P16 was analyzed by semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). Results: 9-cis-RA not only reduced the transcription of Cdk4 but also increased the transcription of P16 in human lung cancer cells. There was no significant difference in the transcription of CyclinD1 between lung cancer tissues and normal lung tissues. The normal lung tissue Cdk4, P16 transcriptional effects were not statistically significant. Conclusion: The mechanism of 9-cis-RA inhibiting the proliferation of lung cancer cells is related to regulating the transcription of Cdk4 and P16.
