汉坦病毒Z37株基因组全序列分析

目的 测定Z37株的全基因组序列 ,了解我国汉坦病毒疫苗生产株分子基础 ,分析其遗传学和抗原性差异等。方法 设计出针对各片段的特异性引物 ,用PCR方法从Z37病毒感染的细胞提取细胞总RNA ,逆转录扩增、产物克隆T载体 ,纯化后测序 ,测定的序列应用DNASTAR软件比较分析。结果 Z37株全基因组由L片段 6 5 30nt、M36 5 1nt、S175 4nt组成 ,分别编码 2 15 1、1133、42 9个氨基酸 ,与同型病毒间同源率高达 95 .2 %~ 99.5 % ,绘出了 3个片段的核苷酸和氨基酸的系统发生树。结论 测定得到了Z37株的全基因组序列 ,为研究疫苗毒株的生物学特性、致病机理等提供了分子基础 Objective To determine the whole genome sequence of Z37 strain and understand the molecular basis of the Hantavirus vaccine production strain in China and analyze its genetic and antigenic differences. Methods Specific primers specific to each fragment were designed. Total RNA was extracted from the cells infected with Z37 by PCR and reverse transcribed and amplified. The T vector was cloned and sequenced. The sequences were analyzed by DNASTAR software. Results The whole genome of Z37 strain consisted of 6 5 30 nt, M36 5 1 nt and S175 4nt L fragments, encoding 215, 1133 and 429 amino acids, respectively. The homology of Z37 was 95.2% ~ 99.5% A phylogenetic tree of 3 fragments of nucleotides and amino acids. Conclusion The whole genome sequence of Z37 strain was obtained, which provided the molecular basis for studying the biological characteristics and pathogenesis of vaccine strain
