激光扫描共聚焦显微镜可对数百 μm厚的组织切片进行共聚焦扫描 ,获得连续的共聚焦平面图像系列 ,通过计算机三维重建算法得到相应组织的立体图像。本文使用 15- 2 0天龄幼年大鼠视皮层脑片的标本 ,在通氧状态下 ,用荧光探针AO染色脑片 ,激光扫描共聚焦显微镜对全脑片不同层面进行共聚焦断层扫描 ,沿纵轴每 2 0 μm扫一次 ,共扫描 16次。再利用总值方式的投影算法对其三维重建 ,得到幼年大鼠视皮层脑片的DNA/RNA分布的立体图像。
