为研究桦褐孔菌菌质多糖对小鼠脾淋巴细胞增殖的影响,采用水提醇沉法提取粗多糖,Sevag法脱蛋白后透析,再经DEAE-Sepharose CL-6B离子交换柱进一步分离纯化,MTT法检测不同浓度的桦褐孔菌菌质多糖对小鼠脾淋巴细胞增殖的影响。试验结果得到3个多糖纯化组分JZP1、JZP2、JZP3;粗多糖(JZPC)、精制多糖(JZPJ)和纯化多糖(JZP3),能够促进淋巴细胞增殖,最大增殖率分别为59.04%(1 000μg/m L),44.58%(20μg/m L),39.76%(20μg/m L)。JZP1和JZP2在1 000μg/m L时抑制淋巴细胞增殖,抑制率分别15.66%和13.25%。结果表明相同浓度下粗多糖对小鼠脾淋巴细胞的增殖作用强于纯化多糖,不同多糖组分表现出不同的免疫学活性,并且其免疫活性与其浓度密切相关。
In order to study the effect of Inonopsis opunas polysaccharide on the proliferation of mouse splenic lymphocytes, crude polysaccharides were extracted by water-alcohol extraction, deproteinized by Sevag method and further purified by DEAE-Sepharose CL-6B ion exchange column MTT assay was used to examine the effects of different concentrations of Inonopsis opunas on the proliferation of mouse splenic lymphocytes. The results showed that three purified polysaccharide components, JZP1, JZP2 and JZP3, could promote the proliferation of lymphocytes with the maximum proliferation rate of 59.04% (1 000 μg / L), the activity of JZPC, JZPJ and JZP3, m L), 44.58% (20 μg / m L), 39.76% (20 μg / m L). JZP1 and JZP2 inhibited lymphocyte proliferation at a dose of 1 000 μg / ml, with inhibition rates of 15.66% and 13.25%, respectively. The results showed that the crude polysaccharide at the same concentration had a stronger effect on the proliferation of mouse splenic lymphocytes than the purified polysaccharide. Different polysaccharides showed different immunological activities, and their immunological activities were closely related to their concentrations.
