目的检测中华骨髓库湖北分库汉族志愿者HLA-DRB1高分辨基因型,了解其基因多态性分布特点。方法采用序列特异性寡核苷酸探针聚合酶链反应(PCR-SSOP)技术对分库1 013名汉族志愿者进行HLA-DRB1高分辨分型。结果共检出HLA-DRB1基因型(低分辨型)12个,检出基因亚型(高分辨型)37个,其中前5位依次为DRB1*0901、DRB1*1501、DRB1*0803、DRB1*1202、DRB1*0401,其基因频率分布与中国台湾慈济脐血库、日本、东欧裔美国人比较,存在统计学差异。结论中华骨髓库湖北分库汉族志愿者HLA-DRB1高分辨基因频率分布相对集中,其基因多态性与中国台湾、日本、东欧裔美国人存在显著性差异。
Objective To detect HLA-DRB1 high-resolution genotypes in Han volunteers of Hubei Marrow Bases in Hubei Province, and to understand the distribution of the polymorphisms. Methods One hundred and thirteen Chinese Han volunteers were screened by HLA-DRB1 using sequence-specific oligonucleotide probe-polymerase chain reaction (PCR-SSOP). Results A total of 12 HLA-DRB1 genotypes (low resolution type) were detected, of which 37 were found subtypes (high resolution type). The top 5 were DRB1 * 0901, DRB1 * 1501, DRB1 * 0803, DRB1 * 1202, DRB1 * 0401, the gene frequency distribution and Taiwan Tzu Chi Umbilical Cord Blood Bank in China, Japan, Eastern European Americans, there is a statistical difference. Conclusion The frequency distribution of HLA-DRB1 high-resolution genes is relatively concentrated in Han volunteers in China Marrow Bank Hubei Branch. The gene polymorphisms of HLA-DRB1 are significantly different from those of Taiwan, Japan and Eastern Europeans in China.
