目的:研究以山嵛酸甘油酯、大豆卵磷脂和胆固醇为油相,泊洛沙姆、聚乙二醇-二硬脂酰磷脂酰乙醇胺和吐温-80作为水相的槲皮素长循环脂质体的制备工艺。方法:采用正交试验法,以纳米脂质体包封率和载药量等作为评价指标,采用多指标综合分析方法优选槲皮素长循环脂质体油相与水相的最佳比例。结果:槲皮素长循环脂质体的最佳制备工艺为:水:油相比例为1.5:1.0,水相之间(泊洛沙姆:PEG2000-DSPE:吐温-80)比例为1:1:1.0,油相之间(山嵛酸甘油酯:大豆卵磷脂:胆固醇)比例为1:2:1,以此制备工艺制得的长循环脂质体包封率和载药量较为理想。结论:该优选工艺制备槲皮素长循环脂质体,方法简易、可行。
OBJECTIVE: To study the long-term quercetin cycle with glyceryl behenate, soy lecithin and cholesterol as the oil phase, poloxamers, polyethylene glycol-distearoylphosphatidylethanolamine and Tween-80 as the water phase Preparation of liposomes. Methods: Orthogonal test was used to evaluate the entrapment efficiency and drug loading of nanoliposomes. The optimal ratio of oily phase and aqueous phase of quercetin long-circulating liposomes was optimized by multi-index comprehensive analysis. Results: The optimal preparation process of quercetin long circulating liposomes was as follows: the ratio of water to oil was 1.5: 1.0, the ratio of water phase to poloxamer: PEG2000-DSPE: Tween-80 was 1: 1: 1.0, the ratio of oil phase (glyceryl behenate: soybean lecithin: cholesterol) is 1: 2: 1, the entrapment efficiency and drug loading of long-circulating liposome prepared by this method are ideal . Conclusion: The optimized process for the preparation of quercetin long circulating liposomes, the method is simple and feasible.
