目的对上海浦东机场蝇类携带新病原菌进行分离鉴定。方法将上海浦东机场口岸捕获的蝇体细菌分离培养至单克隆,使用Vitek2 Compact进行初步鉴定,扩增16S rRNA基因,进行序列比对。使用Mega5,Neighbour-joining方法,构建分子系统发育树。结果从大头金蝇体表分离出一株菌,疑似造成暴发性脓毒症的污蝇解壳杆菌(Wohlfahrtiimonas chitiniclastica)。进化树分析表明为同种菌,16S rRNA相似性为98.66%。结论从大头金蝇体表成功分离到W.chitiniclastica菌的新记录。
Objective To isolate and identify new pathogenic flies in flies in Shanghai Pudong Airport. Methods The flies isolated from Shanghai Pudong Airport Port were isolated and cultured to single clones. Preliminary identification was performed using Vitek2 Compact. The 16S rRNA gene was amplified and sequenced. Use Mega5, Neighbor-joining method to construct molecular phylogenetic tree. As a result, a bacterium suspected to cause fulminant sepsis was isolated from the body surface of the gold-fledged fly, Wohlfahrtiimonas chitiniclastica. Phylogenetic tree analysis showed that the similarity of 16S rRNA was 98.66% for the same strain. Conclusion A new record of W.chitiniclastica was successfully isolated from the body surface of the gold flies.
