目的:观察清热解毒抗风湿方药——热痹康汤对胶原诱导性关节炎(CIA)大鼠滑膜P38/核转录因子-κB(NF-κB)信号通路的影响。方法:将60只SPF级雌性Wistar大鼠随机分为空白对照组,模型对照组,热痹康汤低、中、高剂量组,甲氨蝶呤组,每组10只。除空白对照组外,其余各组大鼠采用弗氏不完全佐剂乳化的牛Ⅱ型胶原蛋白诱导CIA大鼠模型。造模成功后开始给药,模型对照组予生理盐水灌胃,热痹康汤低、中、高剂量组分别给予热痹康汤提取物低、中、高剂量灌胃,甲氨蝶呤组给予甲氨蝶呤腹腔注射,空白对照组大鼠自由饮水和进食。观察不同剂量热痹康汤对CIA大鼠滑膜P38/NF-κB信号通路的影响。结果:热痹康汤能下调CIA大鼠滑膜P38/NF-κB信号通路相关分子P38 MAPK、NF-κB/P65、IκBα的表达。结论:热痹康汤治疗类风湿关节炎的作用与抑制滑膜P38/NF-κB信号通路有关。
Objective: To observe the effect of heat -bijiedu anti-rheumatism prescription-Rebi Decoction on P38 / NF-κB signal pathway in synovial membrane of rats with collagen-induced arthritis (CIA). Methods: Sixty SPF female Wistar rats were randomly divided into blank control group, model control group, Rebukang Decoction low, middle and high dose group and methotrexate group, with 10 rats in each group. Except the blank control group, CIA rat model was induced by bovine type Ⅱ collagen emulsified with Freund’s incomplete adjuvant. After successful modeling, the model rats in the control group were treated with normal saline. The low, medium and high dose of Weibangkang decoction was given low, medium and high dose gavage respectively. The methotrexate group Intraperitoneal injection of methotrexate, blank control group rats free access to water and food. To observe the effects of different doses of Bibi Kang Decoction on synovium P38 / NF-κB signaling pathway in CIA rats. Results: Bweikang Decoction can down-regulate the expression of P38 MAPK, NF-κB / P65 and IκBα in synovial P38 / NF-κB pathway in CIA rats. Conclusion: The effect of Rebukang Decoction on rheumatoid arthritis is related to inhibition of synovial P38 / NF-κB signaling pathway.
