目的探讨不同脂肪酸构成比膳食对大鼠脂联素及其受体表达的影响。方法将72只Wistar大鼠随机分为对照组和5个实验组(A~E),实验组分别设膳食中饱和脂肪酸(SFA)/单不饱和脂肪酸(MUFA)/多不饱和脂肪酸比值(PUFA)(S/M/P)为1:1.7:1.2、1:1:1、2:1:1、1:2:1和1:1:2,喂养12w,测定脂肪组织脂联素(adiponectin,ADPN)、骨骼肌组织脂联素受体1(AdipoR1)、肝脏组织脂联素受体2(AdipoR2)mRNA的表达水平。结果与基础组相比,1:1.7:1.2组和1:1:1组的两种脂肪组织和2:1:1组的皮下脂肪组织脂联素mRNA表达显著降低(P<0.05),其中1:1.7:1.2组下降程度较高(P<0.001)。与1:1.7:1.2组比较,其余各实验组脂联素表达水平均显著升高(P<0.05),以1:1:2组的升高程度最高(P<0.001)。各实验组AdipoR的表达水平与基础对照组并无明显差别(P>0.05)。结论高脂饮食降低脂肪组织脂联素mRNA表达水平,不饱和脂肪酸对促进脂联素基因表达和分泌有明显作用,其中PUFA的作用较MUFA显著。
Objective To investigate the effects of different dietary fatty acids on adiponectin and its receptor expression in rats. Methods Seventy-two Wistar rats were randomly divided into control group and five experimental groups (A to E). The experimental groups were divided into two groups according to the ratio of dietary saturated fatty acid (SFA) / monounsaturated fatty acid (MUFA) / polyunsaturated fatty acid The adiponectin (adiponectin, adiponectin, adiponectin, adiponectin, adiponectin, adiponectin, , ADPN), adiponectin receptor 1 (AdipoR1) and adiponectin receptor 2 (AdipoR2) mRNA in skeletal muscle. Results Compared with the basal group, adiponectin mRNA expression was significantly decreased in the subcutaneous adipose tissue of 1: 1.7: 1.2 group and 1: 1: 1 group and in the subcutaneous adipose tissue of 2: 1: 1 group (P <0.05) 1: 1.7: 1.2 group decreased more (P <0.001). Compared with 1: 1.7: 1.2 group, the levels of adiponectin in the remaining experimental groups were significantly increased (P <0.05), and the highest level was found in 1: 1: 2 group (P <0.001). The expression of AdipoR in each experimental group was not significantly different from that in the control group (P> 0.05). Conclusion High-fat diet can reduce adiponectin mRNA expression in adipose tissue. Unsaturated fatty acid has obvious effect on promoting gene expression and secretion of adiponectin. PUFA is more effective than MUFA.
