目的:研究鬼臼毒素衍生物鬼臼酰肼对K562白血病细胞的抑制增殖、诱导细胞凋亡作用。方法:采用集落形成实验、MTT比色法观察鬼臼酰肼对K562白血病细胞增殖的影响;流式细胞仪等技术手段研究鬼臼酰肼对K562细胞凋亡的影响。结果:鬼臼酰肼能抑制K562白血病细胞增殖,与对照组相比有显著性差异 (P<0. 01),抑制作用与剂量呈显著性正相关。鬼臼酰肼可诱导K562细胞的凋亡,在 5 0μg/ml处理 48h,鬼臼酰肼组凋亡百分率 33 .5%。结论:鬼臼酰肼具有抗肿瘤活性,鬼臼毒素衍生物的不同结构对抗肿瘤活性有一定影响。
OBJECTIVE: To study the inhibitory effect of podophyllotoxin derivative podophyllotrazide on K562 leukemia cells and induce the apoptosis of K562 leukemia cells. Methods: Colony formation assay was used to observe the effects of pisoside-hydrazide on the proliferation of K562 leukemia cells by MTT colorimetric assay. The effects of pisoside-hydrazide on the apoptosis of K562 cells were studied by flow cytometry. Results: Podophyllum hydrazide could inhibit the proliferation of K562 leukemia cells, which was significantly different from the control group (P <0.01). The inhibitory effect was positively correlated with the dosage. Podophyllum hydrazide induced the apoptosis of K562 cells. After treated with 50 μg / ml 48h, the apoptosis rate of psuedohydrazide group was 33.5%. Conclusion: Podophyllum hydrazide has anti-tumor activity. The different structures of podophyllotoxin derivatives have certain influence on anti-tumor activity.
