目的:探讨不同糖皮质激素受体(GR)亚型α、β的蛋白及mRNA在血管瘤患儿瘤组织中水平与糖皮质激素治疗血管瘤疗效的相关性。方法:选取2013年2月~2015年1月来医院就诊并同意接受糖皮质激素治疗的血管瘤患儿56例作为研究对象,治疗前、后均细针穿刺取血管瘤组织,根据治疗效果分为激素治疗有效组和激素治疗无效组。采用酶联免疫法和实时荧光定量法检测两组患儿瘤组织中GRα、GRβ蛋白及mRNA水平。结果:激素治疗有效组患儿瘤组织中GRα蛋白及mRNA水平在治疗前明显高于激素治疗无效组,两组比较差异有统计学意义(P<0.01);GRβ蛋白及mRNA水平在治疗前与激素治疗无效组比较差异无统计学意义(P>0.05)。结论:糖皮质激素治疗血管瘤患儿的疗效与GRα蛋白及mRNA表达水平呈正相关性,与GRβ蛋白及mRNA表达水平无明显相关性,其治疗机制有待于进一步探讨。
Objective: To investigate the relationship between the levels of α and β proteins and mRNAs of different glucocorticoid receptor (GR) subtypes in hemangiomas and the therapeutic effect of glucocorticoid on hemangiomas. Methods: From February 2013 to January 2015, 56 children with hemangiomas who came to the hospital and agreed to receive glucocorticoid therapy were enrolled in this study. Fine-needle aspiration and hemangiomas were taken before and after treatment. According to the therapeutic effect Effective for hormone therapy group and hormone therapy invalid group. Enzyme-linked immunosorbent assay and real-time fluorescence quantitative detection of GRα, GRβ protein and mRNA levels in the two groups of children. Results: The levels of GRαprotein and mRNA in tumor-bearing group were significantly higher than those in hormone-treated group before treatment (P <0.01) Hormone therapy ineffective group was no significant difference (P> 0.05). Conclusion: The therapeutic effect of glucocorticoid on hemangiomas is positively correlated with the expression of GRα protein and mRNA, but not with the expression of GRβ protein and mRNA. The therapeutic mechanism remains to be further explored.
