背景:超声引导下经皮瘤内注入无水乙醇治疗肝癌等局部疗法已在临床得到广泛应用,但仍存在易复发等局限性。目的:探讨复方中药99克星经皮瘤内注射诱导肝癌细胞凋亡的生物效应。设计:随机对照实验研究。地点和对象:实验在福建省超声医学研究所完成,BALB/CA裸鼠,由厦门大学抗癌中心医学实验动物室提供。干预:建立人肝癌SMMC-7721细胞皮下移植与肝内原位移植动物模型各14只。随机分为99克星治疗组14只,无水乙醇与生理盐水各7只,各组中皮下与肝内肿瘤各占50%,每隔5d分别向肿瘤内注入各组治疗药物共4次,20d后处死。主要观察指标:肿瘤中心组织形态,DNA异倍体峰与凋亡峰的出现率。结果:盐水组治疗后肝癌细胞超微结构无明显改变;乙醇组出现以凝固性坏死为主的变化;克星组有92.9%(13/14)见到凋亡细胞或/和凋亡小体。FCMDNA分析结果克星组竟有高达92.9%(13/14)出现肿瘤程序性坏死的凋亡峰,而盐水组却有57.1%(4/7)出现异倍体峰。结论:复方中药99克星具有较强的诱导与促进细胞自发凋亡的作用,似有唤起“自杀基因”的功能。
Background: Ultrasound-guided percutaneous intratumoral injection of absolute alcohol in the treatment of liver cancer and other local therapies have been widely used in clinical practice, but there are still some limitations such as recurrence. Objective: To investigate the biological effects of intraperitoneal injection of compound Chinese herb (99 g) on hepatoma cells. Design: Randomized controlled trial. Location and Subject: The experiment was performed at Fujian Institute of Ultrasound Medicine. BALB / CA nude mice were provided by Medical Laboratory Animal Center, Xiamen University Cancer Center. Intervention: The establishment of human hepatocellular carcinoma SMMC-7721 cell subcutaneous transplantation and orthotopic liver transplantation in 14 animal models. Randomly divided into 99 g star treatment group 14, ethanol and saline 7 each group, subcutaneous and intrahepatic tumors in each group accounted for 50%, respectively, every 5d were injected into the tumor treatment group were 4 times, 20d After the execution. MAIN OUTCOME MEASURES: Tumor center morphology, DNA aneuploidy peak and appearance of apoptotic peak. Results: There was no significant change in the ultrastructure of hepatocellular carcinoma cells after treatment in the saline group. In the alcohol group, coagulation and necrosis were the main changes. 92.9% (13/14) of the nemesis group showed apoptotic cells and / or apoptotic bodies. FCMDNA analysis showed that as many as 92.9% (13/14) of neoplasms in the nemesis group showed apoptotic peak of tumor necrosis while 57.1% (4/7) of the saline group showed aneuploidy peak. CONCLUSION: The 99-g compound Chinese herbal medicine has a strong effect of inducing and promoting spontaneous apoptosis of cells, and seems to evoke the function of “suicide gene”.
