目的:建立心脑康胶囊中葛根素和2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定的RP-HPLC方法。方法:色谱柱:Nova-Pak C_(18)(300mm×3.9mm,4μm),流动相:乙腈-水(20:80),流速:0.8 ml·ml~(-1),检测波长:320nm。结果:葛根素的线性范围为0.31~1.53mg·ml~(-1)(r=0.999 8),平均回收率为98.67%,RSD=1.24%(n=6);2,3,5,4′-四羟基二苯乙烯-2- O-β-D-葡萄糖苷的线性范围为1.21μg·ml~(-1)~9.65μg·ml~(-1)(r=0.999 6),平均回收率为98.43%,RSD=1.02%(n=6)。结论:方法简便、快速、准确,适合该胶囊中葛根素和2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定。
Objective: To establish an RP-HPLC method for the determination of puerarin and 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside in Xinnaokang capsules. Method: Column: Nova-Pak C18 (300mm × 3.9mm, 4μm), mobile phase: acetonitrile-water (20:80), flow rate: 0.8 ml·ml -1, detection wavelength: 320nm. Results: The linear range of puerarin was 0.31~1.53mg·ml -1 (r=0.999 8). The average recovery was 98.67%, RSD was 1.24% (n=6); 2,3,5,4 The linear range of ’-tetrahydroxystilbene-2-O-β-D-glucoside was 1.21μg·ml -1 to 9.65μg·ml -1 (r=0.999 6). The rate was 98.43% with RSD=1.02% (n=6). Conclusion: The method is simple, rapid and accurate. It is suitable for the determination of puerarin and 2,3,5,4′-tetrahydroxystilbene-2-O-β-D-glucoside in this capsule.
