目的:探讨p53凋亡刺激蛋白2(ASPP2)DNA甲基化在老年大鼠肾基脏底膜增厚中的作用和机制。方法:以6月龄大鼠为成年组,18月龄大鼠为老年组,应用过碘酸希夫(PAS)染色,光镜下观察各组大鼠肾脏基底膜改变,qRT-PCR、Western-blot分别检测各组大鼠ASPP2 mRNA及蛋白表达,n MS-PCR法检测ASPP2甲基化水平,qRT-PCR检测DNMT1 mRNA表达改变。结果 :老年大鼠较成年大鼠肾脏基底膜明显增厚,ASPP2 mRNA及蛋白的表达在老年组大鼠中显著增高(P<0.01);与成年组相比,老年组ASPP2 DNA甲基化水平显著下降(P<0.01),且DNMT1 mRNA及蛋白表达也呈下降改变(P<0.01)。结论:ASPP2 DNA甲基化是促进老年大鼠肾脏基底膜增厚的重要机制,DNMT1参与了其调控。
AIM: To investigate the role and mechanism of DNA methylation of p53 apoptosis-stimulating protein 2 (ASPP2) in the thickening of basilar membrane in the kidney of aged rats. Methods: Six-month-old rats were used as the adult group and the 18-month-old rats as the old group. The cells were stained with periodic acid Schiff (PAS) and examined by light microscopy. -blot were used to detect the expression of ASPP2 mRNA and protein in each group. The methylation level of ASPP2 was detected by MS-PCR and the expression of DNMT1 mRNA was detected by qRT-PCR. Results: The expression of ASPP2 mRNA and protein in the aged rats was significantly thicker than that in the aged rats (P <0.01). Compared with the adult group, the levels of ASPP2 DNA methylation (P <0.01), and the expression of DNMT1 mRNA and protein also decreased (P <0.01). Conclusion: ASPP2 DNA methylation is an important mechanism to promote the thickening of basilar membrane in the kidney of aged rats. DNMT1 is involved in its regulation.
