目的通过观察糖酵解通路中相关蛋白M2-型丙酮酸激酶2(PKM2)、哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)、磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(AKT)、缺氧诱导因子1α(HIF-1α)在胃癌患者标本中的表达,以研究PKM2、mTOR、PI3K、AKT、HIF-1α等糖酵解通路中相关蛋白与胃癌之间的相关性。方法采用免疫组织化学法检测89例胃癌患者标本与20例活检正常的胃部组织标本中PKM2、mTOR、PI3K、AKT、HIF-1α等糖酵解通路相关蛋白的表达。结果胃癌组患者组织中PKM2、mTOR的平均光密度值依次为4 423.18±328.77;4 586.08±284.80,均显著高于对照组的相应指标(135.84±15.36,813.55±61.26),差异有统计学意义(P<0.05)。而PI3K、AKT、HIF-1α在二组中差异不明显,无统计学意义(P>0.05)。胃癌患者根据性别、年龄、分期、有无淋巴结转移、分化程度等临床特征进行分组,结果显示胃癌患者在不同性别、年龄及分化程度下PKM2和mTOR的表达率差异无统计学意义(P>0.05);在不同分期、淋巴结转移下PKM2和mTOR的表达率有差异,且具有统计学意义(P<0.05)。结论国人胃癌组织中PKM2和mTOR的相对表达量均显著高于非肿瘤人群,且其表达与胃癌的转移相关,提示PKM2和mTOR与胃癌之间具有相关性。
OBJECTIVE: To investigate the effects of PKM2, mammalian target of rapamycin (mTOR), phosphatidylinositol 3-kinase (PI3K), protein kinase (AKT) and hypoxia-inducible factor-1α (HIF-1α) in gastric cancer patients, so as to study the correlation between the related proteins in gastric cancer such as PKM2, mTOR, PI3K, AKT and HIF- Sex. Methods The expressions of PKM2, mTOR, PI3K, AKT, HIF-1α and other glycoproteins in 89 gastric cancer specimens and 20 biopsied gastric specimens were detected by immunohistochemistry. Results The mean optical density of PKM2 and mTOR in gastric cancer patients were 4223.18 ± 328.77 and 4586.08 ± 284.80, respectively, which were significantly higher than those of the control group (135.84 ± 15.36, 813.55 ± 61.26), the difference was statistically significant (P <0.05). The PI3K, AKT, HIF-1α in the two groups showed no significant difference (P> 0.05). According to gender, age, staging, lymph node metastasis and differentiation, gastric cancer patients were divided into groups according to their clinical features. The results showed that there was no significant difference in the expression rates of PKM2 and mTOR between different gender, age and differentiation (P> 0.05 ). The expression rates of PKM2 and mTOR were significantly different at different stages and lymph node metastasis (P <0.05). Conclusions The relative expression levels of PKM2 and mTOR in gastric cancer tissues in Chinese were significantly higher than those in non-tumorous tissues, and the expression of PKM2 and mTOR correlated with the metastasis of gastric cancer, suggesting that there is a correlation between PKM2 and mTOR and gastric cancer.
