目的探讨促血管生成素-2(Ang-2)对人宫颈癌Hela细胞裸鼠种植瘤血管形成的影响。方法2004年1月至2006年12月在哈尔滨医科大学将实验动物裸鼠分4组:Hank平衡盐溶液组(n=8)、Hela空白对照组(n=9)、Ang-2重组质粒组(n=9)、pcDNA3载体质粒对照组(n=9),比较各组裸鼠成瘤体积、微血管密度(MVD)。结果Ang-2重组质粒组肿瘤体积为(296.72±84.56)mm3,明显小于Hela空白对照组及pcDNA3载体质粒对照组,差异具有统计学意义;Ang-2重组质粒组肿瘤组织中MVD约为12/HP,明显少于Hela空白对照组及pcDNA3载体质粒对照组,差异具有统计学意义。结论Ang-2转染人宫颈癌Hela细胞在裸鼠体内的致瘤力明显降低,肿瘤的体积、肿瘤组织MVD显著低于对照组,表明Ang-2可通过抑制肿瘤血管生成而显著抑制肿瘤生长。
Objective To investigate the effect of angiopoietin-2 (Ang-2) on angiogenesis in human cervical cancer Hela cells implanted in nude mice. Methods From January 2004 to December 2006, nude mice were divided into 4 groups: Hank balanced salt solution group (n = 8), Hela blank control group (n = 9), Ang-2 recombinant plasmid group (n = 9) and pcDNA3 vector plasmid control group (n = 9). The tumor volume and microvessel density (MVD) of nude mice in each group were compared. Results The tumor volume of Ang-2 recombinant plasmid group was (296.72 ± 84.56) mm3, which was significantly lower than that of Hela blank control group and pcDNA3 vector plasmid control group, the difference was statistically significant. The MVD of Ang-2 recombinant plasmid group was about 12 / HP, significantly less than Hela blank control group and pcDNA3 vector plasmid control group, the difference was statistically significant. Conclusion The tumorigenicity of HeLa cells transfected with Ang-2 in human Hela cells was significantly reduced. The volume of tumor and the MVD in tumor tissue were significantly lower than those in control group, indicating that Ang-2 can significantly inhibit tumor growth by inhibiting tumor angiogenesis .
