MicroRNA是一种非编码、小片段、单链的RNA分子,在舌鳞状细胞癌等肿瘤的发生发展中具有重要作用.我们的前期研究显示了miR-29b和miR-195对肿瘤的抑制作用.本研究重点检测了肿瘤细胞和组织中miR-29b和miR-195的表达及其相互关系.针对60对样本中miR-29b和miR-195表达水平的检测结果显示:与癌旁正常组织相比miR-29b和miR-195在肿瘤组织样本中明显呈低表达,而且二者表达水平正向相关.进一步研究显示miR-29b通过抑制其靶基因DNMT3B的表达导致miR-195启动子区CpG岛的甲基化水平降低,从而上调miR-195的表达.直接降低细胞中DMNT3B含量同样导致miR-195启动子区去甲基化水平以及miR-195表达水平的升高,但是同时过表达DMNT3B和miR-29b并不影响miR-195的表达.本研究的结果说明在舌鳞状细胞癌中miR-29b通过直接抑制DMNT3B的表达而上调miR-195的表达.miR-29b和miR-195之间的调控关系可能为舌鳞状细胞癌的治疗提供新的思路.
MicroRNA is a non-coding, small fragment, single-stranded RNA molecule that plays an important role in the development of tumors such as tongue squamous cell carcinoma.Our previous study showed that miR-29b and miR-195 inhibit tumor growth .This study focuses on the detection of miR-29b and miR-195 expression in tumor cells and tissues and their correlation.For the 60 pairs of samples miR-29b and miR-195 expression levels of the test results showed that: adjacent normal tissue phase MiR-29b and miR-195 were significantly lower than that of miR-29b and miR-195 in tumor tissue samples, and the expression of miR-29b was positively correlated with the expression of miR-29b.Further study showed that miR-29b inhibited the expression of DNMT3B, Of DMNT3B decreased the methylation level of miR-195, and increased the expression of miR-195.The direct reduction of DMNT3B content also led to the demethylation of miR-195 promoter region and the increase of miR-195 expression, but over-expression of DMNT3B and miR-29b does not affect the expression of miR-195.The results of this study indicate that miR-29b up-regulates the expression of miR-195 in tongue squamous cell carcinoma by directly inhibiting the expression of DMNT3 B. Between miR-29b and miR-195 The regulatory relationship may be tongue squamous cells The treatment provides new ideas.
