目的:探讨喜炎平注射液对大鼠体内外及人血体外抗血小板聚集作用及其对凝血功能、凝血因子活性的影响。方法:以二磷酸腺苷(ADP)作为诱导剂,测定喜炎平注射液在大鼠体内外及健康人血体外给药对血小板5min之内的最大聚集率;并测定喜炎平注射液对健康人体外全血凝血象和凝血因子活性水平。结果:喜炎平注射液在SD大鼠体内给药剂量400、800mg/kg,体外血药浓度在3.13mg/ml、6.25mg/ml时血小板聚集率与空白对照组比较,可显著性降低血小板5min之内最大聚集率,抑制率与给药剂量呈正相关;健康人体外血药浓度在2.5mg/ml下血小板5min之内最大聚集率与对照组比较,可显著性降低5min之内血小板最大聚集率,抑制率明显提高。健康人的凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)与自身空白对照组比较,时间均有不同程度的延长,具有显著性差异。纤维蛋白(Fi B-C)含量减低、抗凝血酶III(AT-III)活性明显下降。纤维蛋白降解产物(FDP)含量显著性增多。凝血因子II、V、VII、VIII、IX、X、XI、XII的活性与自身空白组比较,均有不同程度的显著降低。结论:喜炎平注射液具有抗血小板聚集作用和抗凝血作用,同时又通过降低AT-III活性来促进凝血的作用,因此,喜炎平注射液对凝血系统既有抗凝也有促凝的双重效果。
