乳猪肝提取物和阿霉素对裸小鼠肾包膜下移植肝癌细胞增殖、分化和凋亡影响的实验研究

目的 :探讨乳猪肝提取物 (LE)和阿霉素 (doxorubicin ,DXR)对裸小鼠肾包膜下移植BEL - 740 2肝癌细胞增殖、分化和凋亡的影响。方法 :应用裸小鼠肾包膜下移植肝癌细胞 ,形态学观察 ,有丝分裂计数 ,超微结构观察和原位DNA末端标记技术。结果 :LE组和DXR组均能明显抑制移植肝癌细胞的生长和促进凋亡。LE对肝癌细胞有丝分裂影响不明显 ,而DXR能抑制其有丝分裂。电镜观察表明 :LE能促进细胞凋亡超微结构的变化 ,表现为细胞固缩、微绒毛消失、核染色质边集固缩和凋亡小体形成 ,而对细胞分化影响不明显。DXR组除有凋亡待征的超微结构变化外 ,还具有促进肝癌细胞分化的作用。结论 :LE能诱发移植于裸小鼠BEL - 740 2肝癌细胞凋亡 ,而对其分化和增殖影响不明显。DXR能促进裸小鼠肝癌细胞凋亡和分化 ,抑制其增殖 Objective : To investigate the effects of sucking liver extract (LE) and doxorubicin (DXR) on the proliferation, differentiation and apoptosis of BEL-7402 hepatoma cells subcutaneously in nude mice. METHODS: Naked mice were subcutaneously transplanted with subcutaneous capsules of hepatoma cells, observed by morphology, mitotic counts, ultrastructure observations and in situ DNA end labeling techniques. RESULTS: Both LE and DXR groups significantly inhibited the growth of transplanted hepatoma cells and promoted apoptosis. LE has no obvious effect on the mitosis of liver cancer cells, but DXR can inhibit its mitosis. Electron microscopy showed that LE could promote the ultrastructural changes of apoptosis, showing pyknosis, disappearance of microvilli, nuclear chromatin condensation, and formation of apoptotic bodies, but had no obvious effect on cell differentiation. In addition to the ultrastructural changes in apoptosis, the DXR group also has the effect of promoting the differentiation of hepatoma cells. Conclusion: LE can induce apoptosis in BEL-7402 hepatoma cells transplanted in nude mice, but the differentiation and proliferation of BEL-7402 hepatoma cells are not obvious. DXR can promote apoptosis and differentiation of hepatoma cells in nude mice and inhibit their proliferation
