目的探讨肼苯哒嗪联合硝酸异山梨醇酯对心力衰竭模型大鼠的影响。方法 40只成年健康雄性大鼠,随机选取10只为正常组,其余进行心力衰竭模型复制,将模型大鼠按数字表法随机分为3组,模型组、实验组、对照组,每组10只。模型组与正常组每日灌胃生理盐水3 m L,对照组予以贝那普利(90mg·kg-1)+比索洛尔氢氯噻嗪片(45 mg·kg-1)灌胃3 m L,实验组予以硝酸异山梨醇酯(800 mg·kg-1)+肼苯哒嗪(225 mg·kg-1),每天灌胃1次3 m L。实验结束后,比较各组大鼠心功能、半胱氨酸(Cys)、同型半胱氨酸(Hcy)、脑钠肽(BNP)、肿瘤坏死因子α(TNF-α)水平及心肌组织形态。结果与模型组比较,实验组与对照组大鼠的BNP、TNF-α水平水平较低,Cys及Hcy水平较高,心功能恢复较好;与对照组比较,实验组大鼠BNP、TNF-α水平水平较低,Cys及Hcy水平较高,心功能恢复较好(均P<0.05)。结论肼苯哒嗪联合硝酸异山梨醇酯能够明显缓解心力衰竭症状对机体的影响,降低血脑钠肽及改善左心室功能、抑制肿瘤坏死因子及同型半胱氨酸的表达。
Objective To investigate the effect of hydralazine combined with isosorbide dinitrate on heart failure model rats. Methods Forty adult healthy male rats were randomly divided into normal group (n = 10) and heart failure model (n = 10). The model rats were randomly divided into three groups according to digital table method: model group, experimental group and control group only. The model group and the normal group were given normal saline 3 m L daily, and the control group was given benazepril (90 mg · kg -1) + bisoprolol hydrochlorothiazide (45 mg · kg -1) The rats were treated with isosorbide dinitrate (800 mg · kg-1) and hydralazine (225 mg · kg-1). After the experiment, cardiac function, levels of cys, Hcy, BNP, TNF-α and myocardial tissue were compared . Results Compared with the model group, the levels of BNP, TNF-α in the experimental group and the control group were lower, the levels of Cys and Hcy were higher, and the cardiac function recovery was better. Compared with the control group, α level was lower, Cys and Hcy levels were higher, heart function recovery is better (all P <0.05). Conclusion Hydralazine combined with isosorbide dinitrate can significantly relieve the effects of heart failure symptoms on the body, decrease blood brain natriuretic peptide and improve left ventricular function, and inhibit tumor necrosis factor and homocysteine expression.
