目的:探讨海藻糖对糖氧剥夺所致SH-SY5Y细胞损伤的保护作用及可能机制。方法:体外培养人SH-SY5Y细胞,建立糖氧剥夺损伤模型。通过乳酸脱氢酶(LDH)释放、锥虫蓝染色来检测细胞存活情况;采用DCFH-DA染色检测细胞内活性氧含量。结果:与对照组相比,海藻糖不仅能够明显抑制缺氧缺糖(OGD)条件下的神经细胞死亡(P<0.05),还能抑制过氧化氢所致SH-SY5Y细胞死亡。结论:海藻糖对糖氧剥夺所致SH-SY5Y细胞损伤的保护作用,其机制可能与其抑制氧化应激反应有关。
Objective: To explore the protective effect of trehalose on SH-SY5Y cell injury induced by glucose-oxygen deprivation and its possible mechanism. Methods: Human SH-SY5Y cells were cultured in vitro to establish a model of glucose-oxygen deprivation injury. Cell viability was measured by lactate dehydrogenase (LDH) release and trypan blue staining. DCFH-DA staining was used to detect intracellular reactive oxygen species. Results: Compared with the control group, trehalose not only significantly inhibited the death of neurons under oxygen and glucose deprivation (OGD) (P <0.05), but also inhibited the death of SH-SY5Y cells induced by hydrogen peroxide. CONCLUSION: The protective effect of trehalose on SH-SY5Y cells induced by glucose-oxygen deprivation may be related to the inhibition of oxidative stress.
