本研究观察了白细胞介素 - 1(IL - 1)、白细胞介素 - 6 (IL - 6 )、单、粒 -细胞集落刺激因子 (GM- CSF)对子宫内膜白血病抑制因子 (LIF)表达的调节 ,探讨围着床期 LIF表达的调节机制。给妊娠 2~ 4 d的小鼠分别腹腔注射不同剂量的 IL - 1、白介素 - 1受体拮抗剂 (IL- 1ra)、IL- 6、GM- CSF,对照组注射 PBS,用免疫印迹法逆转录 -聚合酶链反应进行子宫内膜 LIF蛋白和 m RNA的半定量分析。结果 :对照组 LIF蛋白水平为 39.6 4± 9.95,与对照组比 ,IL - 14μg/kg剂量组明显升高 (P<0 .0 1) ;IL- 6和 GM- CSF 0 .8和 4μg/kg剂量组均明显升高 (分别为 P<0 .0 5,P<0 .0 1)。对照组 L IFm RNA水平为 1.10± 0 .2 1、IL-1、IL- 6和 GM- CSF4 μg/kg剂量组明显高于对照组 (分别为 P<0 .0 5,P<0 .0 5,P<0 .0 1) ,IL - 1ra 2 0μg/kg剂量组与对照组相比明显下降 (P<0 .0 5)。表明 IL- 1、IL - 6、GM-CSF对孕早期小鼠子宫内膜 LIF转录和转录后水平均有上调作用 ,可能是 L IF在围着床期子宫内膜的规律表达的潜在正调节因子
This study investigated the effects of interleukin - 1 (IL - 1), interleukin - 6 (IL - 6), and granulocyte - macrophage colony stimulating factor (GM - CSF) on the expression of endometrial leukemia inhibitory factor To investigate the regulation mechanism of LIF expression around the bed phase. Different doses of IL - 1, IL - 1ra, IL - 6 and GM - CSF were injected intraperitoneally into the mice of 2 ~ 4 days of gestation, and the control group were injected with PBS and reversed by immunoblotting Semiquantitative Analysis of Endometrial LIF Protein and m RNA by Polymerase Chain Reaction. Results: Compared with the control group, the level of LIF protein in the control group was 39.6 4 ± 9.95, which was significantly higher in the IL - 14μg / kg group (P <0.01); IL - 6 and GM - CSF were 0.8 and 4μg / kg dose group were significantly increased (P <0 05, P <0 01). The level of L IFm RNA in the control group was 1.10 ± 0.21, IL-1, IL-6 and GM-CSF4 μg / kg were significantly higher than those in the control group (P <0.05, P < 5, P <0.01). Compared with the control group, IL - 1ra 2 0 μg / kg dose group significantly decreased (P <0.05). The results showed that IL-1, IL-6 and GM-CSF upregulate the transcriptional and post-transcriptional levels of LIF in endometrium of pregnant mice in early pregnancy, which may be the potential positive regulators of the regular expression of LIF in peri-implantation endometrium factor
