【目的】探讨补阳还五汤对脑缺血再灌注大鼠糖原合成酶激酶-3β(GSK3β)m RNA表达与CD63的影响。【方法】将SPF级雄性SD大鼠60只随机分为假手术组,模型组,氢氯吡格雷组,补阳还五汤高、低剂量组,每组12只。氢氯吡格雷组灌胃给予氢氯吡格雷水溶液6.8 mg·kg-1·d-1,补阳还五汤高、低剂量组分别给予补阳还五汤水溶液26、6.5 g·kg-1·d-1,其余各组给予生理盐水;连续给药14 d后,采用右侧大脑中动脉线栓法复制大鼠急性局灶性脑缺血再灌注模型;测定各组大鼠血浆CD63含量与海马组织GSK3βm RNA表达情况。【结果】与假手术组比较,模型组大鼠脑梗死体积显著增加(P<0.05),与模型组比较,补阳还五汤组与氢氯吡格雷组均能显著减少脑梗死体积(P<0.05);与假手术组比较,模型组海马组织GSK3βm RNA表达与血浆中CD63表达均显著升高(P<0.05);与模型组比较,补阳还五汤高、低剂量组与氢氯吡格雷组均能显著抑制海马组织GSK3βm RNA表达与血浆中CD63表达(P<0.05),而补阳还五汤高剂量组抑制作用更明显。【结论】补阳还五汤对脑缺血再灌注大鼠的保护作用可能与下调GSK3βm RNA过表达及抑制CD63表达有关。
【Objective】 To investigate the effect of Buyang Huanwu decoction on glycogen synthase kinase-3β (mrkRNA) expression and CD63 in cerebral ischemia-reperfusion rats. 【Methods】 Sixty SPF male Sprague-Dawley rats were randomly divided into sham operation group, model group, hydrocopagol group and high and low doses of Buyang Huanwu Decoction (n = 12). Hydrochloride-clopidogrel group were given hydrocopagrone aqueous solution 6.8 mg · kg-1 · d-1, Buyang Huanwu Decoction high and low dose groups were given Buyang Huanwu Decoction 26,6.5 g · kg-1 · D-1, and the other groups were given saline; after 14 days of continuous administration, right middle cerebral artery occlusion was used to duplicate acute focal cerebral ischemia-reperfusion model in rats; And hippocampus GSK3βmRNA expression. 【Results】 Compared with the sham operation group, the volume of cerebral infarction in the model group was significantly increased (P <0.05). Compared with the model group, the volume of cerebral infarction was significantly decreased in the Buyang Huanwu Decoction group and the hydrochloropirimine group <0.05). Compared with the sham operation group, the expression of GSK3βmRNA and the expression of CD63 in hippocampus of the model group were significantly increased (P <0.05). Compared with the model group, The pioglitazone group could significantly inhibit the expression of GSK3βmRNA in the hippocampus and the expression of CD63 in the plasma (P <0.05), while the suppressive effect of the BYHWD high dose group was more obvious. 【Conclusion】 Buyang Huanwu Decoction can protect cerebral ischemia-reperfusion rats from over-expression of GSK3βmRNA and down-regulate the expression of CD63.
