建立了一套简便有效的提取方案,可从海带(Laminaria japonica Aresch)中分类提取同时得到褐藻酸钠、褐藻糖胶、褐藻淀粉三类多糖粗品。将含量较高的前二者进一步纯化,乙酸纤维膜电泳和葡聚糖凝胶G-75柱层析的结果表明纯化后的褐藻酸钠、褐藻糖胶是均一的。将二者与市售海藻酸钠试剂的光谱行为和理化特性进行了对比实验,结果表明市售海藻酸钠试剂与褐藻酸钠的紫外和红外图谱特征一致,分子量、特性粘度、电导率及pH值等数值均很接近;而与褐藻糖胶的性质有较大差异。
A simple and effective extraction scheme was established. Three kinds of crude polysaccharides, namely sodium alginate, fucoidan and brown algae, were extracted from Laminaria japonica Aresch. The higher content of the first two further purification, cellulose acetate membrane electrophoresis and Sephadex G-75 column chromatography results show that the purified sodium alginate, fucoidan is uniform. The results of the comparison between the two methods and the spectra and physicochemical properties of the commercial sodium alginate reagent show that the commercial sodium alginate reagent has the same UV and IR spectral characteristics as the sodium alginate and the molecular weight, intrinsic viscosity, conductivity and pH Value and other values are very close; but with the nature of fucoidan are quite different.
