目的 探讨 calponin基因对血管再狭窄的防治作用 ;评价用 infiltrator系统进行基因转染的实用价值 .方法 以p CAGGS质粒为基因载体 ,以脂质体为介导 ,用 infiltrator浸壁球囊导管向兔髂动脉壁内导入人重组 calponin基因 .结果 实验组 (n=6 )基因转染全部成功 . 4wk后转基因组血管内膜增生较轻 ,而对照组 (n=6 )血管内膜明显增生 ,管腔显著变窄 .未观察到与目的基因及转染系统相关的不良反应 .结论 infiltrator导管是活体局部转基因的合适工具 ;用该导管转染 calponin基因可能成为防治血管再狭窄的有效方法 .
Objective To investigate the role of calponin gene in the prevention and treatment of vascular restenosis and to evaluate the practical value of gene transfection with the infiltrator system.Methods The plasmid pCAGGS was used as a vector to mediate liposome and infiltrator In the iliac artery wall, the human recombinant calponin gene was transduced into the wall of the iliac artery.Results The transfection of the experimental group (n = 6) was successful.After 4 weeks, the intimal hyperplasia of the transgenic group was lighter, while the control group (n = 6) Cavity was significantly narrowed and no adverse reaction was observed with the target gene and the transfection system.Conclusion The infiltrator catheter is a suitable tool for local transgene gene transfer.The transfection of calponin gene with this catheter may be an effective way to prevent vascular restenosis.
