目的:建立天麻中对羟基苯甲酸和4,4′-二羟基二苄砜的含量测定方法。方法:采用HPLC法,使用KromasilC18(250mm×4.6mm,5μm)色谱柱,以甲醇-0.1%醋酸水溶液为流动相,梯度洗脱,流速1.0mL.min-1,检测波长240nm,柱温35℃。结果:对羟基苯甲酸和4,4′-二羟基二苄砜进样浓度分别在0.421~8.41μg·mL-1(r=0.9992)和0.650~13.0μg.mL-1(r=0.9999)范围内与峰面积线性关系良好;平均加样回收率(n=9)分别为98.2%和98.7%。结论:采用HPLC法测定天麻中对羟基苯甲酸和4,4′-二羟基二苄砜的含量方法准确可靠,可用于天麻的质量控制。
Objective: To establish a method for the determination of p-hydroxybenzoic acid and 4,4’-dihydroxy dibenzyl sulfone in Gastrodia elata Blume. Methods: The mobile phase was eluted with Kromasil C18 (250mm × 4.6mm, 5μm) using methanol-0.1% aqueous acetic acid as mobile phase with a flow rate of 1.0mL · min-1 and a detection wavelength of 240nm at 35 ℃ . Results: The concentrations of p-hydroxybenzoic acid and 4,4’-dihydroxy dibenzyl sulfone were in the ranges of 0.421-8.41μg · mL-1 (r = 0.9992) and 0.650-13.0μg.mL-1 (r = 0.9999) The linear relationship between peak area and internal area was good. The average recoveries (n = 9) were 98.2% and 98.7% respectively. Conclusion: HPLC method for the determination of p-hydroxybenzoic acid and 4,4’-dihydroxy dibenzyl sulfone is accurate and reliable, and can be used for the quality control of Gastrodia.
