目的考察6种大孔树脂对葶苈子总黄酮成分的分离纯化性能,确定最佳工艺条件。方法以总黄酮吸附量、洗脱率、洗脱物总黄酮含量为指标,采用紫外-可见分光光度法进行含量测定。结果NKA-Ⅱ型树脂对葶苈子总黄酮具有较好的纯化分离能力,其最佳工艺条件为:葶苈子提取物浓度4g/mL(生药量),最大吸附量12.92mg/g,装柱径高比1∶12,吸附流速3mL/min,洗脱剂80%乙醇,洗脱量5倍柱体积,洗脱流速3mL/min。结论NKA-Ⅱ型树脂纯化分离葶苈子总黄酮的最佳工艺稳定高效,可推广用于生产。
Objective To investigate the separation and purification performance of six kinds of macroporous resin on total flavonoids of Fructus Gardeniae, and to determine the optimum conditions. Methods The content of total flavonoids, the elution ratio and the content of total flavonoids in the eluate were used as indicators, and the content was determined by UV-Vis spectrophotometry. Results The NKA-II resin had good purification and separation ability for total flavonoids of Fructus Gardeniae. The optimum conditions were as follows: Fructus Gardeniae extract concentration of 4 g/mL (crude drug), and the maximum adsorption amount was 12.92 mg/g. Column height ratio 1:12, adsorption flow rate 3 mL/min, eluent 80% ethanol, elution volume 5 column volumes, elution flow rate 3 mL/min. Conclusion The optimal process for purification of total flavonoids from Fructus Gardeniae by NKA-II resin is stable and efficient, which can be widely used in production.
