为了进一步探讨山茱萸总甙的抗炎和免疫抑制机制。以ECV3 0 4作为研究HUVEC对象 ,研究了山茱萸总甙对HUVEC表达α4和 β7整合素影响。用rTNF α和rIL 1诱导血管内皮细胞活化 ,模拟血管内皮细胞在炎性条件下的环境 ,采用Cell ELISA方法分析α4整合素、β7整合素的表达 ,结果 ,rTNF α和rIL 1均可诱导血管内皮细胞活化而明显增加α4和 β7整合素的表达 ,地塞米松和山茱萸总甙均可抑制rTNF α和rIL 1诱导ECV3 0 4表达α4和β7整合素 (P <0 0 1)。抑制血管内皮细胞表达α4 β7整合素可能是山茱萸总甙主要抗炎和免疫抑制机制之一。
To further explore the anti-inflammatory and immunosuppressive mechanisms of total hawthorn. Using ECV404 as a HUVEC subject, the effects of total glycosides of Hawthorn on the expression of α4 and β7 integrin in HUVEC were studied. Activation of vascular endothelial cells induced by rTNFα and rIL 1 simulated the environment of vascular endothelial cells under inflammatory conditions. The expression of α4 integrin and β7 integrin was analyzed by Cell ELISA. As a result, both rTNF α and rIL 1 could induce blood vessels. The activation of endothelial cells significantly increased the expression of α4 and β7 integrins. Both dexamethasone and total glucosides of Hawthorn suppressed the expression of α4 and β7 integrins by ECV304 induced by rTNF α and rIL 1 (P <0 01). Inhibiting the expression of α4β7 integrin in vascular endothelial cells may be one of the main anti-inflammatory and immunosuppressive mechanisms of total glycosides in Hawthorn.
