拟松材线虫组织蛋白酶基因Bmcath1是一个在松材线虫与拟松材线虫中差异表达的基因,为了解该基因的生物学功能,对Bmcath1基因进行了生物信息学分析。笔者使用BioEdit、SignalP 4.1、BLASTP、MEGA 4.0等软件对Bmcath1基因进行了生物信息学分析,发现Bmcath1 cDNA序列全长1 294 bp,含有一个长度为1 158 bp的开放阅读框。由Bmcath1基因编码的蛋白质CATH1含有386个氨基酸,分子质量为4238 ku,等电点为8.52。蛋白质CATH1存在一个Peptidase_C1A subfamily功能域结构,在不同物种上的氨基酸序列多样性丰富。采用Real-Time PCR方法对不同培养方式获得的拟松材线虫中Bmcath1基因的表达量进行检测,发现该基因在分离自黑松的拟松材线虫中高量表达,据此推测拟松材线虫Bmcath1基因可能在线虫处于松树体内生长、繁殖时发挥更为重要的作用。
The Bmcath1 gene of B.subtilis was a differentially expressed gene in pine wood nematode and pine wood nematode. To understand the biological function of this gene, the Bmcath1 gene was analyzed by bioinformatics. Bioinformatics analysis of Bmcath1 gene using BioEdit, SignalP 4.1, BLASTP, MEGA 4.0 and other softwares showed that Bmcath1 cDNA was 1 294 bp in length and contained an open reading frame of 1 158 bp in length. The protein CATH1 encoded by the Bmcath1 gene contains 386 amino acids with a molecular mass of 4238 ku and an isoelectric point of 8.52. The protein CATH1 has a Peptidase_C1A subfamily domain structure and rich diversity of amino acid sequences in different species. Real-Time PCR method was used to detect the expression level of Bmcath1 in different culture methods to obtain the Bmcath1 gene. The results showed that Bmcath1 gene was highly expressed in the pine wood nematode isolated from Pinus thunbergii, suggesting that Bmcath1 Genes may play a more important role in pine growth and reproduction.
