红细胞生成素(Epo)是一重要的造血因子,能刺激红系前体细胞的增殖、分化及成熟。目前,重组人红细胞生成素(rHuEpo)已广泛地应用于临床,其生物学、药理学研究也越来越深入。本文就有关进展作一综述。 生物学特性 rHuEpo为一种糖蛋白,有165个氨基酸残基,分子结构中60%为蛋白质、40%为糖。蛋白链的分子重量为18.4kd、糖基的分子重量约为30.4kd。分子结构特点为β片层基础上的螺旋结构,有2个二硫键维持结构的立体形状,周围有4个糖基区。rHuEpo的生物学活性依赖糖基,若除去糖基中的唾液酸则失去活性。
Epo is an important hematopoietic factor that stimulates the proliferation, differentiation and maturation of erythroid precursors. At present, recombinant human erythropoietin (rHuEpo) has been widely used in clinical, its biological, pharmacological research is also more and more in-depth. This article reviews the progress. Biological Features rHuEpo is a glycoprotein with 165 amino acid residues, 60% of the protein in the molecular structure and 40% sugar. The molecular weight of the protein chain is 18.4kd and the molecular weight of the glycosyl group is about 30.4kd. The molecular structure is characterized by a β-sheet helical structure with two disulfide bonds that maintain the three-dimensional shape of the structure, surrounded by four glycosylated domains. The biological activity of rHuEpo is dependent on the glycosyl group and is lost if the sialic acid in the glycosyl group is removed.
