目的探讨小檗碱对小鼠艾氏腹腔积液肿瘤细胞的影响,并阐述其作用机制。方法将50只小鼠随机分为空白对照组、环磷酰胺(CTX)组、小檗碱3组(不同剂量),每组各10只。每只鼠注入0.5 ml艾氏腹腔积液肿瘤细胞,CTX组予CTX 5 mg/kg,小檗碱组分别予小檗碱5、10、15 mg/kg腹腔注射。计算小鼠生存时间及生命延长率,并测定巨噬细胞吞噬功能、肿瘤坏死因子α(TNF-α)细胞杀伤率和自然杀伤(NK)细胞活性。结果存活时间空白对照组、CTX组、小檗碱不同剂量(5、10、15 mg/kg)组分别为(12.2±3.3)d、(17.8±4.6)d、(15.4±2.8)d、(17.5±3.0)d、(19.8±2.7)d,除空白对照组外,各组生命延长率分别为39.7%、29.2%、42.8%、59.7%。用药组与空白对照组比较差异均有统计学意义(P<0.05);CTX组与小檗碱组比较差异无统计学意义(P>0.05);小檗碱组随给药剂量增加存活时间有所延长,用药剂量与小鼠存活时间呈正相关,差异有统计学意义(P<0.05)。用药组巨噬细胞吞噬功能、TNF-α细胞杀伤率、NK细胞活性与空白对照组比较差异有统计学意义(P<0.05,P<0.01);CTX组与小檗碱组比较差异无统计学意义(P>0.05);小檗碱组间巨噬细胞吞噬功能比较差异无统计学意义(P>0.05)。TNF-α细胞杀伤率随小檗碱剂量增大呈增高趋势,小檗碱15 mg/kg组NK细胞的活性有所降低,但差异无统计学意义(P>0.05)。结论小檗碱对小鼠肿瘤具有一定的抑制和促溶作用,可明显延长小鼠存活时间。
Objective To investigate the effect of berberine on Ehrlich ascites tumor cells in mice and its mechanism of action. Methods Fifty mice were randomly divided into blank control group, cyclophosphamide (CTX) group and berberine group (10 doses) at 10 different doses. Each mouse injected with 0.5 ml Ehrlich’s ascites tumor cells, CTX 5 mg / kg CTX group, berberine group berberine 5,10,15 mg / kg intraperitoneal injection. The survival time and life extension rate of mice were calculated. Phagocytosis of macrophages, cytotoxicity of tumor necrosis factor α (TNF-α) and natural killer (NK) cell activity were measured. Results The survival time of blank control group, CTX group and berberine group were (12.2 ± 3.3) d, (17.8 ± 4.6) d and (15.4 ± 2.8) d 17.5 ± 3.0) d and (19.8 ± 2.7) d, respectively. Except for the blank control group, the life prolongation rates in each group were 39.7%, 29.2%, 42.8% and 59.7%, respectively. The difference between the two groups was statistically significant (P <0.05); there was no significant difference between the CTX group and the berberine group (P> 0.05); the berberine group increased with the dose of Prolonged, the dose of drug was positively correlated with the survival time of mice, the difference was statistically significant (P <0.05). The phagocytosis of macrophages, the killing rate of TNF-α cells and NK cell activity in the treatment group were significantly different from those in the blank control group (P <0.05, P <0.01); there was no significant difference between the CTX group and the berberine group (P> 0.05). There was no significant difference in the phagocytic function of macrophages between berberine groups (P> 0.05). The cytotoxicity of TNF-α increased with the dose of berberine. The activity of NK cells in berberine 15 mg / kg group was decreased, but the difference was not statistically significant (P> 0.05). Conclusion Berberine has some inhibitory and anti-dissolubility effects on tumor in mice, which can significantly prolong the survival time of mice.
