目的:对秀珍菇子实体多糖中的未知糖组分进行分析鉴定。方法:秀珍菇子实体经热水提取分离纯化得到纯多糖PGPS1。PGPS1经三氟乙酸水解后用硼氢化钠还原,乙酰化后以单糖标准品为对照,采用气相色谱(GC)和气相色谱-质谱(GC-MS)联用分析,进一步采用13C NMR DEPT-135和2D NMR(1H-1H COSY谱、TOCSY谱、HMQC谱、HMBC谱和NOESY谱)分析该未知糖组分。结果:GC分析和GC-MS联用发现PGPS1中含有葡萄糖、甘露糖、岩藻糖和一种3(4)-O-甲基-己糖,核磁结果解析证明该己糖为3-O-甲基-半乳糖。结论:PGPS1中含有3-O-甲基-半乳糖。
Objective: To analyze and identify the unknown sugar components in the polysaccharides of fruiting bodies of Pleurotus ostreatus. Methods: The fruiting bodies of Pleurotus ostreatus were isolated and purified by hot water to obtain pure polysaccharide PGPS1. PGPS1 was hydrolyzed with trifluoroacetic acid and then reduced with sodium borohydride. After acetylation, the monosaccharide standard was used as a control. Gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) were used to analyze PGPS1 and 13C NMR DEPT- 135 and 2D NMR (1H-1H COZY spectrum, TOCSY spectrum, HMQC spectrum, HMBC spectrum and NOESY spectrum). Results: Glucose, mannose, fucose and 3 (4) -O-methyl-hexose were found in PGPS1 by GC and GC-MS. The results of NMR analysis showed that the hexose was 3-O- Methyl-galactose. Conclusion: PGPS1 contains 3-O-methyl-galactose.
