荷叶碱是荷叶中的一种阿朴啡型生物碱,为荷叶中主要的降脂活性成分。为能够充足的利用和提取荷叶中的荷叶碱,在酶辅助酸性乙醇提取荷叶生物碱的方法中,基于Box-Behnken响应曲面分析方法对酶解过程进行优化。研究酶添加量、酶解时间、酶解pH和酶解温度对荷叶生物碱提取效果的影响。结果表明,酶解最佳工艺条件为酶添加量0.14%、酶解时间4 h、酶解pH4.5和酶解温度45℃。在此条件下荷叶生物碱的提取率稳定,且与理论值接近,说明该响应曲面模型具有实际应用价值。
Nuciferine is a type of aporphine alkaloids in lotus leaf, which is the main lipid-lowering active ingredient in lotus leaf. In order to fully utilize and extract the nuciferine in the lotus leaf, the enzymatic hydrolysis process was optimized based on the Box-Behnken response surface methodology in the extraction of lotus leaf alkaloids by enzyme-assisted acidic ethanol. The effects of enzyme dosage, enzymolysis time, pH and temperature on the extraction of alkaloids from lotus leaf were studied. The results showed that the optimum conditions of enzymatic hydrolysis were 0.14% of enzyme dosage, 4 hours of enzymolysis time, pH 4.5 and enzymolysis temperature of 45 ℃. Under these conditions, the extraction rate of lotus leaf alkaloids is stable and close to the theoretical value, indicating that the response surface model has practical value.
