观察了催产素反义肽抗体(antibodyofantisensepeptideoxytocin,简称AS- OT-Ab)对催产素受体(OTR)的识别特性。该反义肽是按照牛催产素(OT)和它的结合蛋白NeurophysinI(NPI)氨基端前十二个氨基酸(包括OT[1-9])编码DNA的互补序列固相合成的,简称AS-OT。AS-OT与甲状腺球蛋白联结后,制备兔抗AS-OT多克隆抗体(AS-OT-Ab),间接酶联免疫吸附测定(ELISA)证明,AS-OT-Ab能与催产素受体(OTR)相互结合,而这一结合能被OT所阻断,显示了OT与其反义肽抗体的相似性。
The recognition of oxytocin receptor (OTR) by the antibody of antisense peptideoxytocin (AS-OT-Ab) was observed. The antisense peptide is synthesized in solid phase according to the complement of bovine oxytocin (OT) and the DNA encoding the first twelve amino acids (including OT [1-9]) of the amino terminus of its binding protein Neurophysin I (NPI) OT. AS-OT and thyroglobulin were conjugated to prepare rabbit polyclonal anti-AS-OT antibody (AS-OT-Ab). The indirect enzyme-linked immunosorbent assay (ELISA) demonstrated that AS- OTR) bind to each other, and this combination can be OT blocked, showing the OT and its antisense peptide antibody similarity.
